Phenylethylamine (PEA)
PEA is a trace amine your brain makes naturally from L-phenylalanine — the same molecule associated with the lift people describe after intense exercise or during early infatuation. It acts on TAAR1 (trace-amine-associated receptor 1) and promotes the release of dopamine and noradrenaline, briefly raising catecholamine tone in reward-related circuits.
PEA's effect is fast and short. It's metabolised quickly by the enzyme MAO-B, which is exactly why its action is acute rather than lingering — a "spark," not a sustained drug-like effect. In Formula 02 it sits alongside compounds that support the same dopamine pathway from other angles, so the role of PEA is to initiate the dopaminergic lift the rest of the stack then supports.
A quick, clean lift in the first few minutes — more interested, more forward-leaning, less flat. Brief by design.
Kanna (Zembrin®, Sceletium tortuosum)
Zembrin® is a standardised extract of the South African plant Sceletium tortuosum, used traditionally for centuries to relieve stress and lift mood. Pharmacologically it's characterised as a dual mechanism: serotonin (5-HT) reuptake inhibition + PDE4 inhibition. The PDE4 action raises cAMP → CREB signalling — the same intracellular cascade tied to mood and cognitive flexibility.
In a randomized, double-blind, placebo-controlled trial, 25 mg of Zembrin® daily was well tolerated over 3 months. A pharmaco-fMRI study found a single 25 mg dose reduced reactivity of the amygdala (the brain's threat-detector) to fearful faces, and altered amygdala–hypothalamus coupling — a neural signature consistent with reduced anxiety load. A separate crossover RCT reported improvements in cognitive set-flexibility and executive function. 25 mg is the exact dose used in the clinical work — Formula 02 doses it on-label, not below.
The floor comes up. Less reactive, less on-edge — a steadier baseline that makes it easier to engage rather than brace.
Alpha-GPC (Alpha-Glycerylphosphorylcholine)
Alpha-GPC is a highly bioavailable choline source. Choline crosses the blood–brain barrier and feeds acetylcholine synthesis — the neurotransmitter behind attention, learning and mental clarity. In 02 it provides the cholinergic underlay beneath the dopaminergic drive, so motivation has clarity behind it rather than scattered restlessness.
Acute supplementation with Alpha-GPC has been shown to improve cognitive performance (Stroop test) in healthy young adults, and meta-analysis supports its cognitive benefit in clinical populations.

L-Tyrosine
L-Tyrosine is the direct dietary precursor of dopamine and noradrenaline. It's converted by tyrosine hydroxylase to L-DOPA, then to dopamine. Supplemental tyrosine gives the synthesis pathway more raw material — most useful precisely when catecholamines are being depleted faster than they're made, i.e. under stress or heavy cognitive load.
Reviews of placebo-controlled studies show tyrosine can support working-memory and cognitive performance under demanding or stressful conditions, by replenishing catecholamine substrate.

Mucuna pruriens (15% L-DOPA)
Mucuna pruriens (velvet bean) is a natural source of L-DOPA — the direct, immediate precursor of dopamine and the one molecule in the chain that needs no rate-limited conversion step before becoming dopamine. Standardised to 15% L-DOPA, the 40 mg dose delivers roughly 6 mg of plant-derived L-DOPA, feeding the dopamine pathway downstream of where L-Tyrosine enters it.
Where L-Tyrosine supplies raw material at the top of the pathway, Mucuna supplies L-DOPA directly — a complementary, two-point approach to supporting dopamine synthesis. Mucuna seed extract also carries antioxidant and neuroprotective constituents beyond its L-DOPA content.
The "want to" returns — a sense of drive and engagement rather than effortful pushing.
Rhodiola rosea (3% rosavins)
Rhodiola is an adaptogen — a class of botanicals studied for buffering the body's stress response. It's associated with modulation of the HPA (stress) axis and cortisol response, and with reduced mental and physical fatigue, helping protect the dopaminergic state from being eroded by stress.
Phase-III RCTs of standardised Rhodiola extract (SHR-5) in stress-related fatigue reported anti-fatigue effects, improved concentration, and reduced cortisol response to waking stress. A systematic review of RCTs covers its use for physical and mental fatigue.
Stress sits lighter; the mood lift holds up better against a demanding day.
Saffron (affron®, Crocus sativus)
Saffron® is a standardised saffron extract (standardised to ≥3.5% Lepticrosalides — safranal + crocins). Saffron's mood-supporting actions are linked to its effects on serotonin availability and antioxidant activity in mood-relevant circuits.
In randomized, double-blind, placebo-controlled trials, 28 mg/day of affron® improved mood and reduced stress markers in adults with low mood; saffron at 28–30 mg has repeatedly shown mood benefit across trials.

BioPerine® (Piperine 95%)
BioPerine® is a standardised black-pepper (Piper nigrum) extract — 95% piperine — included as a bioavailability enhancer. Piperine increases gastrointestinal absorption of co-administered nutrients by stimulating amino-acid transporters, modifying the gut epithelial membrane, and slowing the enzymes that metabolise actives before they reach circulation.
In the landmark human study, piperine increased curcumin bioavailability dramatically; BioPerine® has separately been shown to raise plasma levels of co-administered nutrients (β-carotene, B6, selenium, CoQ10). In Formula 02 its job is to help the rest of the stack actually get absorbed — a force-multiplier on every other compound. At 15 mg it's a generous, deliberate inclusion.

The Cofactor Layer
Dopamine isn't made from precursors alone — the enzymes that build it need vitamin cofactors. Formula 02 supplies them in active, ready-to-use forms so the synthesis chain isn't bottlenecked:
- Vitamin B6 as P-5-P — the active cofactor for AADC, the enzyme that converts L-DOPA → dopamine.
- Vitamin C — cofactor for dopamine β-hydroxylase, which converts dopamine → noradrenaline.
- Niacin & B12 (Methylcobalamin) — support energy metabolism and methylation around monoamine turnover.
